There is no list of seven medications that are proven to cause severe dementia and profound brain damage in people when taken as prescribed. However, some medications can impair memory and thinking, especially with long-term use, high doses, or in older adults. In some cases, these effects are reversible after the medication is stopped, while in others, they may contribute to an increased risk of cognitive decline.
Here are seven medication classes commonly associated with cognitive impairment:
- Anticholinergic medications

- Long-term use of strong anticholinergics has been associated with a higher risk of dementia in some observational studies, but this does not prove they directly cause dementia.
- Benzodiazepines

- Sedative-hypnotics (“Z-drugs”)

- Opioid pain medications

Do these medications cause “profound brain damage”?
For most of these medications, no. They typically affect brain function rather than causing permanent structural brain damage when used appropriately. Severe brain injury is more likely to result from events such as:

- Severe drug overdose leading to respiratory arrest.
- Stroke.
- Serious infections affecting the brain.
